In this presentation we presented an innovative integrated system designed to automate the tangential flow filtration (TFF) process based on inline concentration measurement. This system employs in-line variable pathlength spectroscopy to acquire concentration measurements, enabling real-time monitoring and control of the process and ensuring desired results. This approach is robust and immune to the variability issues that frequently arise in mass balance-only systems. The system’s automated process control capabilities are demonstrated across a range of final concentration endpoints, indicating superior performance to traditional ultrafiltration/diafiltration (UF/DF) methods.

This new Process Analytical Technology (PAT)-driven TFF system represents a significant advancement in UF/DF technology with distinct advantages over existing methods.
