Fast Electrophoretic Analysis of Genome Libraries in the Next-Generation Sequencing Workflow

Over the last few years, novel sequencing platforms have greatly lowered the cost-per-base of DNA and RNA sequencing. In particular, lower costs coupled with significant improvements in data quality have enabled sequencing projects that may have previously been too expensive to complete.

As a result, the demand for high-quality libraries has escalated. Laboratories have invested significant effort in improving library preparation and workflows through introducing new equipment and automation. In particular, the need for quality control (QC) technology that is compatible with established work flows and in-place next-gen sequencing instrumentation has assumed increasing importance.

In this webinar, use and application of the Agilent 2200 TapeStation system at the Wellcome Trust Centre for Human Genetics in Oxford will be described.

The Agilent 2200 TapeStation system, a benchtop system compatible with 16-tube strip vials and 96-well plates, allows accelerated sample analysis, yielding qualitative information within the next-gen workflow. The instrument provides flexibility, ease of use, and an accessible solution for sample and library QC compatible with high- and low-throughput projects with minimal hands-on time, displaying fully analyzed results in approximately one minute per sample.

Comparisons of results from various stages of the Agilent Sure Select target-enrichment Illumina NGS protocol to the current “gold standard” system – the Agilent 2100 Bioanalyzer system will also be presented. By providing a range of ScreenTape consumables with standard and high sensitivities along with a tailored analysis package, the Agilent 2200 TapeStation system can QC fragmented DNA, whole genome libraries, and target-enriched libraries, providing descriptive analyses at each stage.
