Tangential flow filtration is a common processing step for concentration and diafiltration (buffer exchange) operations in the downstream processing of biopharmaceutical products. Using a pre-sanitized, single use TFF membrane makes it possible to reduce the total number of process steps and thus reduce labor by 50% or more. By eliminating non-value added cleaning and flushing steps, buffer and water usage is reduced by 75% or more. In addition to the cost savings realized from the reduced labor and buffer usage, single use TFF can increase productivity and throughput, by >45% in many cases. The process economics for single use TFF are very favorable when comparing the costs of reusable vs, single use TFF in biopharmaceutical applications.

In downstream processing, many TFF systems are traditionally operated as clean and reuse systems. A typical reusable TFF system will involve ten major process steps (installation, CIP, flush, normalized water permeability (NWP), equilibrate, process, post-use CIP, flush, post-use NWP, and storage). Using a pre-sanitized, disposable TFF membrane enables the strategy for single use to reduce the number of process steps from ten down to three (installation, equilibrate, process). While there will be significant cost savings realized from the reduced labor, reduced buffer usage and reduced validation requirements, the major benefit for utilizing a single use strategy for single use TFF is an increase in productivity and throughput. The benefits of single use TFF are especially useful for clinical manufacturing and contract manufacturing operations because the scale of operation and frequency of change over in these operations fit well into a favorable economic model.

Capabilities

TangenX manufactures ultrafiltration and microfiltration membranes based on the chemistry of modified polyethersufone (mPES). We offer two membrane chemistries with specific characteristics to optimize your TFF process. Our ProStream™ membrane is a low protein binding membrane with neutral charge. ProStream mPES is well suited for compatibility with viruses, oligonucleotides, liposomes, and recombinant proteins.

Our HyStream™ membrane is a highly hydrophilic mPES membrane with excellent product recovery and particularly well performing with monoclonal antibodies. Both ProStream and HyStream membranes are available in our single use and reusable formats. Our Sius Single Use TFF cassettes and Novaset™ reusable format TFF cassettes are available in a wide range of pore sizes and offered in a complete range of membrane surface areas to support use in process development, pilot-scale and cGMP process-scale TFF operations.
